摘    要:
目的观察anti-miR-155反义寡核苷酸(AMOs)对肺腺癌A549细胞增殖的影响。方法 A549细胞分为对照组和AMOs处理组,采用AMOs抑制A549细胞内miR-155的活性,液闪计数仪测定[3^H]-TdR掺入量,MTT法测定细胞增殖抑制率,流式细胞仪测定细胞周期。结果与对照组相比,AMOs显著减少A549细胞[3^H]-TdR掺入量,随着浓度从10nmol/L逐渐增加至100nmol/L,A549细胞[3^H]-TdR掺入量亦随之减少。MTT法测定细胞增殖抑制率结果显示,与对照组相比,AMOs显著抑制A549细胞的增殖。流式细胞术检测结果显示AMOs使G0/G1细胞比例显著增加,G2/M期细胞比例显著减少。结论采用AMOs抑制A549细胞内高水平表达miR-155的活性后,可显著抑制A549细胞的增殖。

Abstract:
Objective To determine the effect of anti-miR-155 oligonucleotides (AMOs) on proliferation of human lung adenocarcinoma cell line A549. Methods A549 cells were divided into two groups: control group and AMOs group. AMOs were used to inhibit the activity of miR-155. [3^H]-TdR incorporation was measured by liquid scintillation counting. MTT assay was used to observe the inhibition percentage of AMOs on the proliferation of A549 cells. Cell cycle was measured by flow cytometry. Results AMOs significantly decreased the [3^H]-TdR incorporation in a dose-dependent manner. With the increment of concentration of AMOs, the inhibition percentage was significantly increased. AMOs significantly decreased the cell ratios of G2/M phase and S phase, simultaneously increased the cell ratio of G0/G1 phase. Conclusion AMOs inhibits the activity of miR-155 highly expressed in A549 cells, as a result, significantly inhibits the proliferation of A549 cells.
