滋肾丸对去卵巢骨质疏松雌鼠的骨组织形态学的改善作用及机制的实验研究

目的:探讨补肾中药滋肾丸对去卵巢大鼠骨质疏松的骨形态学的改善作用及机制研究。方法:将66只SD大鼠随机分为6组,每组11只,空白组做假手术,其余5组做卵巢切除术,术后2周开始给药,用药12周后处死。处死后取股骨远端1/3部分及胫骨近端1/3进行骨组织形态学研究,取血测定血清雌二醇、碱性磷酸酶含量。结果:模型组的骨量明显减少,骨组织退化,出现骨小梁数目减少,厚度变薄,面积百分率下降;血清碱性磷酸酶升高,血清雌二醇下降。中药滋肾丸大、中、小剂量及尼尔雌醇均能对上述指标有所改善,差异具有统计学意义(P<0.05)。结论:补肾中药滋肾丸对去卵巢骨质疏松大鼠的骨组织形态学有改善作用,使其骨量增加,骨结构改善。其疗效与其升高血清雌二醇,降低碱性磷酸酶,从而降低骨高转换率有关,表明补肾中药滋肾丸对骨质疏松有一定的治疗作用。

Experiment research on effect and the machanism of tectology of bone of female SD rat after taking kidney - reinforcing Chinese herb ZiShen Wan

Objective:To investigate the effect and the mechanism of tectology of bone after kidney-reinforcing Chinese herb Zishenwan applied to gelding osteoporosis female rat.Methods:Female SD rat(n=66,15 weeks old) were randomly assigned to 6 groups of 11 each,the control group were shamly operated;the other groups were performed ovariectomy on both sides.Medicines were given for 12 weeks,beginning from 2 weeks after surgery,then they are killed.Take the outlying 1/3 quarters of thighbone and close 1/3 quarters of shinbone,investigate the tectology of bone;collect the blood,determin E_2 and ALP.Results:lumber number of bone significantly reduce,the bone organization degraded,the number of trabecular reduced,the thickness become thin,the area percent declined.The level of ALP increased and the level E_2 of decreased significantly in model group.Zishenwan group including big,middle,small dosage group,can antagonize these changes which has the statistics meaning,(P<0.05).Conclusions:Kidney-reinforcing Chinese herb Zishenwan can improve the tectology of Bone of gelding osteoporosis female rat,the mechanism has relationship with its raising the level of E_2 in blood and reduce of level of ALP to depress the hyper bone turnover.
