GDF-5对大鼠肌腱细胞及腱鞘滑膜细胞增殖及凋亡的影响

目的 观察生长分化因子-5(GDF-5)对体外培养的大鼠肌腱细胞及腱鞘滑膜细胞增殖的影响,为进一步研究在体内局部添加GDF-5以促进损伤肌腱愈合及改善外源性愈合所致粘连打下基础.方法 MTT法测定不同浓度的GDF-5在体外对肌腱及腱鞘滑膜细胞增殖率的影响;流式细胞仪测定不同浓度的GDF-5在体外对肌腱及腱鞘滑膜细胞凋亡率的影响.结果 GDF-5可以明显促进肌腱细胞的增殖,而对腱鞘滑膜细胞的增殖率的作用较弱.对两种细胞的凋亡无明显影响.结论 添加一定浓度的GDF-5后肌腱细胞增殖率较腱鞘滑膜细胞提高更为明显,而凋亡并未发生改变.

Effect of GDF-5 on Proliferation and Apoptosis of Tendon Cells and Sheath Cells of Rats

Objeetive To explore the effects of growth differentiation factor-5(GDF-5) on proliferation of in vitro cultured tendon cells and sheath cells of rats,and lay the foundation for our further research on enhancing healing of hurt sheath by GDF-5 and alleviating adhesions by endogenous healing in vivo.Methods The proliferation rate of tendon cells and sheath cells was measured by using MTT assay.The apoptosis rate of tendon cells and sheath cells was detected by using flow cytometry.Results GDF-5 could obviously promote the proliferation of tendon cells,but had little impact on the proliferation of sheath cells.GDF-5 had no effects on the apoptosis of both tendon cells and sheath cells.Conclusion Addition of GDF-5 can enhance the proliferation rate of tendon cells more significantly than sheath cells.
