The Change of Hypericin Content in Hypericum ascyron

OBJECTIVE To determine whether there is hypericin in Hypericum ascyron and changes of hypericin content. METHODS Hypericin was extracted by 80% ethanol and the contents were determined by high-performance liquid chromatography. The samples were separated on a SHIMADZU Shim-pack VP-ODS C₁₈ column (4.6 mm×150 mm，5 μmand detected at 590 nm by a SPD-20A UV detector. The mobile phase consisted of methanol-0.006 mol·L-1 disodium hydrogen phosphate (87.5∶12.5, v/v, adjusted pH to 6.5 with phosphoric acid at the flow rate of 1.0 mL·min-1. The column temperature was at 30 ℃. RESULTS Hypericum ascyron contains hypericin and the contents were changed with the seasons. The hypericin contents in stems and leaves reached peak in May and trough in August. The hypericin content in flowers did not change with season. The contents were different in various kinds of organs in the same month and it was high in flowers, medium in leaves , low in stems. CONCLUSION Hypericum ascyron contains hypericin, and hypericin content changes with the seasons and in different organs.
