大蒜素对大鼠急性脑缺血再灌注损伤保护作用的研究

摘    要:目的 研究大蒜素 (allicin)对局灶性脑缺血再灌注损伤的保护作用及机制。方法 采用线栓法制备大鼠大脑中动脉缺血再灌注模型 ,于缺血 3h再灌注 2 4h后测定大鼠脑组织中超氧化物歧化酶 (SOD)和髓过氧化物酶 (MPO)的活性、丙二醛 (MDA)、谷胱甘肽 (GSH)及血浆中一氧化氮 (NO)含量。结果 大蒜素 10、2 0mg·kg-1能将SOD活性从2 0 5 87± 16 5 2分别提高到 2 5 9 77± 19 6 5和 2 84 5 4±2 1 79,将MDA从 38 6 9± 0 10分别降至 2 8 6 8± 1 37和2 0 97± 0 4 4 ,将GSH含量从 2 8 31± 0 6 4分别提高到36 19± 1 12和 4 7 96± 0 4 4 ,将NO含量从 2 9 7± 0 4 9分别降至 2 7 38± 1 4 6和 2 3 0 0± 0 71,将MPO从 0 2 9±3 0 6× 10 -2 分别降至 0 2 6± 2 13× 10 -2 和 0 2 3± 7 5 2×10 -2 。结论 大蒜素对大鼠局灶性脑缺血再灌注损伤有明显保护作用 ,其机制可能与提高抗氧化酶活性 ,抗脂质过氧化损伤及抗炎作用有关

Abstract:Aim To study the protective effects of allicin on acute cerebral ischemia reperfusion injury. Methods The model of cerebral ishemia-3 h/reperfusion-24 h was induced by middle cerebral artery occlusion(MCAO) in SD rats. Allicin(10,20 mg·kg -1) was administered once daily in rats:at 0 h of reperfusion. After 24 h reperfusion, the content of malondialdehyde(MDA),the activities of superoxide dismutase(SOD),GSH and myeloperoxidase(MPO) in brain tissue,the content of NO in plasma were measured. Results Compared with vehicle group, the activity of SOD was increased from 205.87±16.52 to 259.77±19.65 and 284.54±21.79 by allicin 10,20 mg·kg -1; the MDA content was decreased from 38.69±0.10 to 28.68±1.37 and 20.97±0.44 respectively; the GSH content was increased from 28.31±0.64 to 36.19±1.12 and 47.96±0.44 respectively; the NO content was decreased from 29.7±0.49 to 27.38±1.46 and 23.00±0.71 respectively; the MPO content was decreased from 0.29±3.06×10 -2 to 0.26±2.13×10 -2 and 0.23±7.52×10 -2 respectively. Conclusion The protective effects of allicin on acute cerebral ischemia reperfusion injury may be related to increasing antioxidase activities,decreasing lipid peroxidative damage and inhibiting inflammation reaction.
