摘    要:目的 研究熊果苷对体外培养的正常人表皮黑素细胞的作用效应。方法 利用碱性成纤维细胞生长因子为主要有丝分裂原建立正常人表皮黑素细胞培养系,MTT法测定熊果苷对黑素细胞活力的影响,以左旋多巴为底物测定酪氨酸酶活性,比色法测定黑素含量,并与曲酸及维生素C磷酸酯的结果比较。结果 熊果苷对体外培养的正常人黑素细胞酪氨酸酶活性呈浓度依赖性抑制,可显减少黑素生成量,对黑素细胞活力影响很小。结论 熊果苷对体外培养的人黑素细胞的酪氨酸酶活性及黑素生成有显抑制作用,且细胞毒性很低。

Abstract:AIM To study the effects of arbutin on cultured human melanocytes. METHODS Basic fibroblast growth factor (bFGF) was employed to establish the culture of normal human melanocytes. After arbutin was added into cultured human melanocytes, the cell viability, tyrosinase activity and melanin content were measured by MTT assay, utilization of L Dopa as the substrate, and colorimetric analysis, respectively. Furthermore, the effects of arbutin on the melanocytes were compared with that of kojic acid and Ascorbic acid 2 phosphate. RESULTS Arbutin showed a concentration dependent reduction in tyrosinase activity of human melanocyte. Melanin content was reduced significantly by arbutin, which has little influence on melanocytes viability. CONCLUSION Arbutin can significantly inhibit the tyrosinase activity and melanogenesis of cultured human melanocytes with low cytotoxicity.
