| 摘 要： | 目的 探讨五味子乙素(Sch B)抗H2O2诱导PC12细胞凋亡的影响及机制.方法 在H2O2诱导PC12细胞凋亡模型的基础上,采用MTT比色法分析细胞存活率; Hochest33258荧光染色法和流式细胞仪检测细胞凋亡情况;DCFH-DA法检测细胞内活性氧(ROS)水平;Real Time PCR和Western印迹检测p53基因mRNA和蛋白的表达.结果 不同剂量Sch B处理PC12细胞48 h后,可剂量依赖性对抗H2O2引起的细胞凋亡,提高细胞存活率,减轻H2O2导致的细胞核固缩、聚集和碎裂,降低细胞内ROS的含量,抑制p53基因mRNA和蛋白的表达.结论 Sch B可剂量依赖性对抗H2O2诱导的PC12细胞凋亡,其机制可能与其抗氧化、下调促凋亡基因p53的表达等有关.
