熊果酸对过氧化氢诱导大鼠心肌细胞氧化损伤的保护作用

摘    要:目的观察熊果酸(ursolic acid,UA)对过氧化氢(H2O2)诱导大鼠心肌细胞氧化应激损伤模型的保护作用,并探讨其作用机制。方法冷消化法原代培养SD大鼠心肌细胞,3 d后将细胞随机分为5组:正常对照组、H2O2模型组、UA低(2.5μM)、中(5μM)、高(10μM)浓度组。UA预处理心肌细胞后,加入H2O2作用,分别检测心肌细胞存活率、LDH、CAT、SOD的变化,ROS的水平。结果 UA低、中、高浓度组对比模型组OD值明显降低,LDH含量明显下降,CAT、SOD活性升高,ROS水平下降,且P0.05。结论 UA对大鼠心肌细胞氧化损伤具有保护作用,可减轻氧自由基对心肌细胞的氧化损伤。

Abstract:Objective To investigate the effect of ursolic acid on the rat myocardial cell with hydrogen peroxide ( H2 O2 ) induced oxidative stress damage model and explore its action mechanism .Methods SD rat myocardial cells were cultivation primitived with cold digestion method ,then were divided to five groups after three days:normal control group,H2O2 model group,low UA(including 2.5μM),medium(including 5μM), high concentration group(including 10 μM).After myocardial cells were preprocessed with UA and H2O2, detected the survival rate of myocardial cell and the changes of LDH ,CAT,SOD,the level of ROS.Results Compared with the model group ,US groups's OD value with low ,medium and high concentration were lowered significantly ,the levels of LDH were decreased significantly ,the activity of CAT and SOD were increased ,the levels of ROS were decreased .ALL of them were P<0.05 .Conclusion UA could protect the rat myocardial cell out of the oxidative damage ,and it could reduced the Oxidative damage to rat myocardial cell by oxygen free radicals .
