| 摘 要： | 目的建立C57BL/6小鼠成肌细胞体外培养、纯化和鉴定的方法。方法采用Ⅱ型胶原酶和胰蛋白酶消化分离新生C57BL/6小鼠的成肌细胞,用Percoll分离液纯化细胞,在含15%胎牛血清的DMEM培养基中进行成肌细胞培养,倒置相差显微镜下观察细胞生长情况,Desmin抗体鉴定成肌细胞。结果经过Ⅱ型胶原酶和胰蛋白酶消化分离,Percoll纯化的成肌细胞纯度较高,Desmin染色95%以上的细胞胞浆染色阳性。结论采用Percoll纯化、国产胎牛血清培养,可以获得高纯度的成肌细胞。

| Abstract: | Objective To establish the methods for culture,identification and purification of C57BL/6 mouse myoblasts in vitro.Methods The mouse skeletal muscle was digested with typeⅡ Collagenase and Trypsin and the isolated cell suspension was purified by Percoll.Then mouse myoblasts were cultured in medium contained 15% FCS,The cells observed by invert microscope,the myoblasts were identified by Desmin immunochemical staining.Results At least 95% myoblasts were harvested by using this way,up to 95% myoblasts showed positive Desmin staining.Conclusion Myoblasts could be purified by Percoll and cultured with DMEM cultured liquid contained FCS made in our country,in high quantity. |
