大鼠急性胰腺炎凋亡相关蛋白Fas、FasL、Bcl-2的表达及意义

摘    要:
目的 :探讨凋亡调控基因Fas、FasL、Bcl 2在大鼠急性胰腺炎中的表达情况及其与胰腺炎严重程度的关系。方法 :通过胰胆管逆行注射不同浓度的脱氧胆酸钠 ,制作急性水肿性胰腺炎 (AEP)和急性坏死性胰腺炎(ANP)模型。用免疫组化法检测Fas、FasL、Bcl 2蛋白的表达。结果 :急性胰腺炎时 ,Fas蛋白主要表达在胰腺腺泡细胞胞膜 ,AEP组和ANP组均明显高于对照组 (P <0 .0 5 ) ,且AEP组高于ANP组 (P <0 .0 5 ) ;FasL在各组腺泡细胞胞浆中均有不同程度表达 ;Bcl 2蛋白表达在胰腺导管细胞胞浆 ,对照组Bcl 2蛋白表达弱 ,AEP组和ANP组表达明显增强 ,ANP组高于AEP组 ,统计学处理两组间无明显差异 ,但均显著高于对照组 (P <0 .0 5 )。结论 :Fas/FasL系统在大鼠的急性胰腺炎过程中可能参与诱导胰腺腺泡细胞凋亡 ,减轻胰腺炎症反应程度 ;而Bcl 2基因可能参与防止导管细胞凋亡而促进其增生并且形成导管管状复合体

Abstract:
Objective: To study expression of apoptosis regulated genes Fas,FasL and Bcl 2 in experimental acute pancreatitis(AP) and its correlation with the severity of the disease in SD rats.Methods: The models of acute edematous pancreatitis(AEP) and acute necrotizing pancreatitis(ANP) were established by retrograde injection of different concentration of sodium deoxycholate into the common bile pancreatic duct.The expression of Fas,FasL and Bcl 2 protein was detected by immunohistochemical method.Results: In AP group,Fas was mainly expressed in the cytomemberous of acini cells,the expression in both AEP and ANP group was significantly higher than that in the control group( P <0.05),and that in the AEP group was higher than in the ANP group( P < 0.05 ). Immuno staining revealed that moderate to strong FasL immunoreactivity was present in the cytoplasm of the acini of all groups. The expression of Bcl 2 protein occurred in the cytoplasm of duct cells in both AEP and ANP groups which was significantly higher than that in the control group ( P <0.05).Though its expression in the ANP group was higher than it in the AEP group,the different was not statistically significant.Conclusion: The Fas/FasL system may contribute to acini cells apoptosis and reduce the severity of AP.Bcl 2 gen expression in duct cells may resist to apoptosis,and might promote cell proliferation,thus to form tubular complexes.
