摘    要:目的:探索血浆组织型纤溶酶原活化因子(t-PA)在青光眼视网膜中的表达变化、可能的作用以及脑源性神经营养因子(BDNF)对其的影响。方法:采用前房灌注法建立兔急性高眼压模型,在玻璃体内注射BDNF或BSS液,用免疫组织化学法检测视网膜t-PA的蛋白表达与定位,用Westernblot分析检测视网膜t-PA的蛋白表达。结果:t-PA与β-actin灰度比值,正常对照组为(57.95±3.79)%、BSS组为(89.36±3.59)%、BDNF组为(66.66±2.16)%;BSS组与正常对照组和BDNF组相比,t-PA表达明显升高(P<0.05)。高眼压后视网膜上t-PA的免疫组化染色明显增强,神经纤维层、RGCs层呈强阳性染色。结论:t-PA与青光眼的病理过程有相关性,BDNF可能部分通过抑制t-PA的表达发挥神经元保护作用。

Abstract:Objective:To examine the expression change and potential role of tissue plasminogen activation(t-PA) in the glauco-matous retina and to investigate the possible mechanism of effects of BDNF on the expression of t-PA.Methods:Acute intraocular hypertension models were established by irrigating the anterior chambers.The rabbits received the intravitreal injection with BBS or BDNF.Western blot and immunohistochemistry were used to check the change of t-PA.Results:The brightness ratio of the strap of t-PA vs β-actin from the normal control group,Group BSS to Group BDNF was(57.95±3.79)%,(89.36±3.59)%,(66.66±2.16)% respec-tively;The level of t-PA is significantly higher in the BBS group than the BDNF group and the control group(P<0.05).The immunohis-tochemistry labeling of t-PA increased significantly after hypertension especially in nerve fiber layers and ganglion cells.Conclusions: The up-regulation of t-PA expression may correlate with pathologic process of glaucoma.BDNF may partly protect the retina,especial-ly the retina ganglion cells through inhibiting the expression of t-PA.
