基因芯片快速检测常见水中致病菌的初步应用研究

摘    要:目的:利用基因芯片技术,尝试对从实际样品中分离的几种常见致病菌进行快速检测鉴定。方法:通过PCR扩增检测靶基因,采用基因芯片与扩增产物在一定条件下进行杂交,杂交结果通过ScanArray 3000芯片扫描仪读取并与标准杂交图谱比较,从而判定样品中细菌的种属。结果:对分离的20株细菌进行了基因芯片的杂交检测,并用传统方法对这些菌株进行了鉴定,基因芯片检测结果与传统方法鉴定结果的一致性为95%(19/20)。结论:基因芯片技术检测水中常见致病菌具有快速、准确、易于操作等优越性,值得推广应用。

Abstract:Objective:To detect and identify rapidly several common pathogens in water by gene chip technology. Methods: The target gene is amplified by PCR and hybridized with gene chip. The hybridized result was read by ScanArray 3000 chip scanner and compared with standard hybridization map to determine the species of bacteria in the sample.Result: 20 isolated bacteria were detected by gene chip and also detected by traditional method, the coherence of the two methods was 95%(19/20). Conclusion: Gene chip technology to detect common pathogens in water is a rapid, accurate, and easy to manipulate that it is worth propagation.
