摘    要:目的建立肾石通颗粒中槲皮素的含量测定方法。方法采用高效液相色谱法KromasilC18色谱柱(250mm×4.6mm,5μm),以甲醇-0.4%磷酸溶液(50:50)为流动相,检测波长:370nm,流速:1.0ml·min^-1,柱温:25℃。供试品溶液制备采用甲醇回流提取1h,聚酰胺柱层析中用70%乙醇提取效果较好。结果槲皮素在5.20~20.80mg·L^-1的浓度范围内峰面积与量呈良好的线性关系,回归方程为:A=3.858×10^5×C+8.094×10^3(R=0.99998,n=7);平均加样回收率可达98.9%,(RSD=0.40%,n=9)。结论本法具有专属、可靠、灵敏度高、干扰小等特点,可作为肾石通颗粒中槲皮素的含量测定方法。

Abstract:Aim To estalish a method for determination of the content of quercetin in shenshitong granule. Methods HPLC method was used. The sample was separated by Kromasil C18 column (250 mm×4.6 mm,5 μm). The mobile phase was methanol-0.4% phosphoric acid (50 : 50). The detection wavelength was 370 nm. Flow rate was 1.0 ml · min^-1. the column temperature was at 25℃. Results The linear range of quercetin is between 5.20 mg · L^-1 and 20.80 mg · L^-1. The average recovery is 98.9% (RSD = 0.40% ,n = 9). Conclusions The method is sensitive, rapid and accurate. It is suitable for determining shenshitong grainule.
