摘    要:目的建立测定血清万古霉素质量浓度的高效液相色谱法..方法以HypersilC,s柱(250mm×4.6mill,10μm)为色谱柱,流动相为乙腈一磷酸二氢钾缓冲液(10:90),流速为0.8mL/min,检测波长为236nm,以10%硫酸锌(ZnSO4)为沉淀血清蛋白,离心后取上清液直接进样测定。结果万古霉素的低、中、高(1.50,30.82,150.45mg/L)3种质量浓度的平均相对回收率分别为99.33%,98.94%和99.70%,日内及日间精密度的RSD均小于5%(n=5);分析方法的最低检测质量浓度为0.5mg/L;线性范围为2.o~150mg/L,回归方程为C=0.06108A+0.0003845,r=0.9999。结论该方法灵敏、准确、简单、快速,适用于血清万古霉素血药浓度的监测和药物代谢动力学研究。

Abstract:Objective To estsblish a HPLC method for the determination of serum mass concentration of vancomycin. Methods Van- comycin was analyzed by the RP-HPLC system with Hypersil Cs column(250 mm *4.6 mm,10 μm) and the mobile phase of ace- tonitrile-potassium dihydrogen phosphate (10:90).The flow rate was 0.8 mL/min and the UV detection wavelength was 236 nm. With 10% ZnSO4 as the precipitation serum albumin, the supernatant after centrifugation was taken for detection by direct sample in- jection. Results The average recovery rate of vancomycin in low,medium and high concentrations (1.50,30.82,150.45 mg/L) were 99.33% ,98.94% and 99.70% ,respectively. The inter-day and intra-day relative standard deviations(RSD) were less than 5% (n =5). The minimum detectable concentration of serum vancomycin was 0.5 mg/L. The linear range was 2.0- 150 mg/L. The regress equa- tions was C = 0. 061 08 A + 0. 000 384 5, r = 0. 999 9. Conclusion This method shows to be sensitive,accurate, simple, rapid, and suitable for monitoring the serum concentration of vancomycin and its pharmacokinetic study.
