注射用丹参多酚酸盐对动脉粥样硬化大鼠的作用和机制探讨

摘    要:目的:探讨注射用丹参多酚酸盐对动脉粥样硬化(AS)大鼠的治疗作用和可能机制。方法:取10只正常大鼠为对照组,其余大鼠采用腹腔注射维生素D3+高脂饲料喂养诱导AS模型,将造模成功的27只大鼠随机分为模型组、丹参多酚酸盐组和阿托伐他汀组,并给予相应的药物腹腔注射4周。第16周HE染色观察主动脉病理变化,检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、氧化型低密度脂蛋白(ox-LDL)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)、白介素-10(IL-10)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)和主动脉中单核细胞趋化蛋白(MCP-1)、血管细胞黏附分子(VCAM-1)、基质金属蛋白酶-9(MMP-9)的含量。结果:与对照组比较,模型组大鼠主动脉斑块形成,脂质沉积,管腔狭窄,血清TC、TG、LDL-C、ox-LDL、TNF-α、IL-1β、IL-6、MDA和主动脉MCP-1、VCAM-1、MMP-9含量明显升高(P<0.05),HDL-C、IL-10、SOD、GSH-Px含量明显降低(P<0.05)。与模型组比较,丹参多酚酸盐组和阿托伐他汀组大鼠主动脉无明显斑块形成,内膜增厚,血清TC、TG、LDL-C、ox-LDL、TNF-α、IL-1β、IL-6、MDA和主动脉MCP-1、VCAM-1、MMP-9含量明显降低(P<0.05),HDL-C、IL-10、SOD、GSH-Px含量明显升高(P<0.05)。结论:注射用丹参多酚酸盐可以通过调节脂代谢,抑制炎症和抗氧化损伤,改善动脉粥样硬化。

Abstract:Objective:To investigate the therapeutic effect and the possible mechanism of Zhusheyong Danshen Duofensuan Yan on atherosclerosis(AS)rats.Methods:Ten normal rats were taken as the control group.The remaining rats were fed with vitamin D3 and high-fat diet to induce the AS model.27 successfully modeled rats were randomly divided into the model group,Danshen Duofensuan Yan group and atorvastatin group,and the corresponding drugs were injected intraperitoneally for 4 weeks.At the 16 thweek,HE staining was used to observe the pathological changes of the aorta.The levels of total cholesterol(TC),triglycerides(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),and oxidized form Low-density lipoprotein(ox-LDL),tumor necrosis factor-α(TNF-α),interleukin-1(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malondialdehyde(MDA)in serum and monocyte chemotactic protein(MCP-1),vascular cell adhesion molecule(VCAM-1),matrix metalloproteinase-9(MMP-9)in aorta were detected.Results:Compared with the control group,the aortic plaque formation,lipid deposition,lumen stenosis,the contents of TC,TG,LDL-C,ox-LDL,TNF-α,IL-1β,IL-6,MDA in serum and MCP-1,VCAM-1,MMP-9 in aorta of the model group were significantly increased(P<0.05),while the contents of HDL-C,IL-10,SOD,GSH-Px were significantly decreased(P<0.05).Compared with the model group,there was no obvious plaque formation in the aorta of Danshen Duofensuan Yan group and atorvastatin group,and the intima was thickened.The content of TC,TG,LDL-C,ox-LDL,TNF-α,IL-1β,IL-6,MDA in serum and MCP-1,VCAM-1,MMP-9 in aorta were significantly decreased(P<0.05),while the content of HDL-C,IL-10,SOD,GSH-Px was significantly increased(P<0.05).Conclusion:Zhusheyong Danshen Duofensuan Yan can improve atherosclerosis by regulating lipid metabolism,inhibiting inflammation and anti-oxidative damage.
