uPA和MMP-9在乳腺浸润性导管癌中的表达及其相关性

摘    要:目的探讨尿激酶型纤溶酶原激活剂(uPA)和基质金属蛋白酶(MMP-9)在乳腺癌及肿瘤周围组织中的表达与乳腺癌侵袭、转移的关系。方法应用免疫组化Envision法检测50例乳腺癌及20例癌旁组织中uPA和MMP-9蛋白的表达情况,并分析其相关性及与临床病理特征进行比较。结果乳腺癌组织中uPA的阳性表达率为60%(30/50),MMP-9的阳性表达率为66%(33/50),显著高于癌旁组织(P〈0.01),在乳腺癌组织中uPA、MMP-9蛋白表达之间存在显著正相关(r=0.630,P〈0.01)。uPA、MMP-9表达与淋巴结转移、临床分期显著相关(P〈0.05),与患者的年龄、肿瘤大小无明显相关性(P〈0.05)。结论乳腺癌组织中存在uPA、MMP-9的高表达;uPA蛋白可通过诱导MMP-9蛋白的袁达上调,促进乳腺癌侵袭、转移。

Relationship between the expression of uPA and MMP-9 in invasive ductal carcinoma of breast

Abstract:Objective To study the expression and correlation of urokinase-type plasminogen activator(uPA) and matrix metaUopmteinase-9 (MMP-9) in invasive ductal carcinoma of breast. Methods Expression of uPA and MMP-9 was detected in 50 cases of breast carcinoma tissue and 20 patients with normal breast tissues adjacent to tumor was examined by immunohistochemi- cal staining. Results The positive expression rates of uPA and MMP-9 in breast carcinoma were 60% ,66% respeetively, show- ing a significant difference between breast carcinoma and normal breast tissues adjacent to tumor(P〈0. 01 ). The expression of uPA was positively correlated with expression of MMP-9 in breast carcinoma (r = 0. 630, P〈 0. 01 ). The expression of uPA and MMP-9 was significantly correlated with lymphatic metastasis and surgical-pathologic staging(P〈0. 05) ,but not significantly cor- related with age and tumor size (P〉0.05). Conclusion The overexpression of uPA and MMP-9 is found in breast carcinoma. To induce uPA overexpression may be one of pathways of MMP-9 that enhances invasion and contributes to the infiltration and metas- tasis in human breast carcinoma.
