亚硒酸钠对枸杞幼苗磷脂氢谷胱甘肽过氧化物酶活性和硒含量的影响

摘    要:目的探讨亚硒酸钠(Na2SeO3)对宁夏枸杞幼苗磷脂氢谷胱甘肽过氧化物酶(PHG-PX)活性和硒(Se)含量的影响。方法将75株宁夏枸杞幼苗随机分为对照组、Na2SeO30.5 mg.kg-1组(每千克土壤施用Na2SeO30.5 mg)和Na2SeO32.0 mg.kg-1组(每千克土壤施用Na2SeO32.0 mg),每组25株。分别于处理后24 h、48 h、72 h及96 h测定PHG-PX活性和Se含量。Se含量采用国际标准GB12399-90进行测定,PHG-PX活性以烟酰胺腺嘌呤二核苷酸磷酸(NADPH)的减少速度来表示。结果Na2SeO30.5 mg.kg-1组处理幼苗48 h和Na2SeO32.0 mg.kg-1组处理24 h后,PHG-PX活性高于对照组(P<0.05)。Na2SeO32.0 mg.kg-1组处理后各时间点PHG-PX活性均高于Na2SeO30.5 mg.kg-1组,48 h后达到显著水平(P<0.05),96 h后达到极显著水平(P<0.01)。0.5 mg.kg-1和2.0 mg.kg-1Na2SeO3处理后各时间点枸杞幼苗Se元素的含量均高于对照组(P<0.01)。Na2SeO32.0 mg.kg-1组处理后各时间点枸杞幼苗的Se含量均高于Na2SeO30.5 mg.kg-1组(P<0.01)。结论外源Se的摄入能够提高枸杞幼苗体内Se元素的含量和PHG-PX活性,从而提高枸杞幼苗的营养价值和保健价值。

Abstract:Objective To explore the effects of Na2SeO3 on the content of selenium and the activity of phospholipid hydropexide glutathione peroxidase (PHG-PX) of Lycium barbarum seedlings. Methods 75 Lycium barbarum seedlings were randomly divided into control group, 0. 5 mg·kg^-1 Na2SeO3 group (0. 5 mg Na2SeO3 were used per kilogram soil ), 2.0 mg·kg^-1 Na2SeO3 group(2.0 mg Na2SeO3 were used per kilogram soil) ,25 in each group. The content of selenium and the activity of PHG-PX were determined at 24 h,48 h,72 h,96 h after treatment. The content of selenium was determined by international standard GB12399-90. The activity of PHG-PX expressed with the decreasing velocity of nicotinamide adenine dinucleotide phosphate (NADPH). Results The activity of PHG-PX at 48 h after treated in 0.5mg·kg^-1 Na2SeO3 group and 24 h after treated in 2.0 mg·kg^-1 Na2SeO3 group were significantly higher than that in control group(P 〈0.05). The activity of PHG-PX in 2.0 mg·kg^-1 Na2SeO3 group was higher than that of 0.5 mg·kg^-1 Na2SeO3 group and the difference achieved statietic significance after 48 h ( P 〈 0.05 ) ;The difference was more obviously after 96h ( P 〈 0.01 ). The content of selenium in 0.5 mg·kg^-1 Na2 SeO3 group and 2.0 mg·kg^-1 Na2 SeO3 group were significantly higher than that in control group ( P 〈 0. 01). The content of selenium in 2.0 mg·kg^-1 Na2SeO3 group was higher than that in 0.5 mg·kg^-1 Na2SeO3 group(P 〈 0. 01 ). Condusion Absorption of exterior selenium can increase the content of internal selenium and the activity of PHG-PX of Lycium barbarum seedlings. So it can enhance the nutritive value and hygienical function of Lycium barbarum seedlings.
