砷对小鼠肝脏脂质过氧化的影响及Vit E干预作用的研究

摘    要:目的:研究不同水平的砷对小鼠肝脏组织脂质过氧化的影响 ,并观察维生素 E(Vit E)对砷毒作用的干预作用。方法 :昆明种小鼠按两因素三水平析因实验设计分为 9组 ,不同实验组的动物通过喂饲法给予三氧化二砷和 Vit E复制亚慢性砷中毒动物模型。三氧化二砷的分组剂量按照其对小鼠经口半数致死量 (L D50 )进行分组 ,Vit E按照我国青少年每日推荐量进行分组。 2个月后研究砷对小鼠肝脏组织中超氧化物歧化酶 (SOD)活力、谷胱甘肽过氧化物酶 (GSH - Px)活力及丙二醛 (MDA )含量的影响以及给于 Vit E处理后的干预作用。结果:染毒组肝脏组织中 SOD、 GSH- Px活力低于正常对照组 (P <0 .0 5 ) ,MDA含量高于正常对照组 (P <0 .0 5 ) ;给于 Vit E处理后 ,动物肝脏中 SOD、GSH- Px活力增强 ,MDA含量降低 (P <0 .0 5 )。结论:砷能引起小鼠肝脏组织中氧化与抗氧化系统失衡 ,Vit E可拮抗砷对小鼠肝脏氧化水平与抗氧化能力的影响。

Abstract:Objective: To study the influence of arsenic and the antagonistic action of vitamin E on lipid peroxidation in the liver of mice. Methods: Kunming mice were divided into nine groups according to the factorial experiment design of two factors and three levels. The mice of different groups were fed with both various amounts of arsenic in drinking water and vitamin E for two months to produce the animal model of subchronic arsenic poisoning. The mice were fed the different dose of arsenic the LD_ 50 and the dose of vitamin E according to the youngsters RDA (recommended dietary allowance). The activity of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px) and the contents of malondiasldehyde (MDA) in the liver of mice were measured by test kit. Results: (1) The SOD and GSH-Px contents were lower ( P < 0.05 ),while the MDA contents were higher ( P <0.05) in arsenic groups. Correspondingly,the SOD and GSH-Px contents were higher ( P <0.05),while the MDA contents were lower ( P <0.05) in the groups added with various dose of vitamin E (abbreviated by Vit E As). Conclusions: (1) Lipid peroxidation (LPO) and the anti-lipid peroxidation might play a vital role in the mechanism of arsenism. (2) There was an interaction between arsenic and vitamin E. Vitamin E has an antagonistic action against the arsenism through mataining the balance of the lipid peroxidation and anti-lipid peroxidation system.
