摘 要: | 目的研究饱和氢气生理盐水对大鼠心肌缺血再灌注模型梗死质量百分比、血流动力学变化及血清肿瘤坏死因子(TNF-α)表达的影响。方法 90只大鼠随机分为3组:G1组,假手术组(n=30);G2组,缺血再灌注(I/R)组(n=30);G3组,缺血再灌注用饱和氢气生理盐水处理组(n=30)。结扎大鼠冠状动脉前降支(LAD)30 min,松扎再灌注24 h,制备心肌缺血再灌注损伤模型,再灌注24 h后测量心肌梗死质量百分比、血流动力学及TNF-α的基因表达情况。结果与G1组相比,再灌注24 h后G2、G3组左心室收缩压(LVSP)、等容收缩期左心室压力上升的最大速率[+(dp/dt)max]、等容舒张期左心室压力下降的最大速率[-(dp/dt)max]显著下降(P0.01),左心室舒张末压(LVEDP)显著增加(P0.01),TNF-α表达明显上调(P0.01)。与G2组相比,再灌注24 h后G3组心肌梗死质量百分比明显减小(P0.01),LVSP、+(dp/dt)max、-(dp/dt)max显著增加(P0.01),LVEDP显著下降(P0.01),TNF-α表达明显下调(P0.01)。结论饱和氢气生理盐水能有效减小梗死质量百分比,改善血流动力学参数,其机制可能与其降低炎性反应相关。

Abstract: | Objective To study the possible effects of saturated hydrogen-rich saline on the rat model of myocardial ischemiareperfusion(I/R) injury. Methods Ninety rats were equally randomized and assigned to three groups:sham-operated group(n = 30),I/R group(n = 30) and I/R treated with H2 saline group(n = 30). Myocardial I/R model was induced by occlusion of the left anterior descending(LAD) coronary artery for 30 min,then the tube was removed for myocardial reperfusion for 24 h. After reperfusion for24 h,the infarct size was measured,the hemodynamic change was detected and the gene expression of TNF-α was detected. Results The infarct quality in G3 was significantly smaller than that in G2(P 〈0.01). LVSP,LVEDP,+(dP/dt) max and(dP/dt) max in each group were significantly different(P 〈0.01),respectively. The expression of TNF-α increased in G2 and G3 compared with G1(P 〈0.01),whereas the expression of TNF-α decreased significantly in G3 compared with G2(P 〈0.01). Conclusion Saturated hydrogen-rich saline has the effect of cardioprotection by reducing the infarct quality and improving hemodynamic parameters probably through the mechanism of anti-inflammatory reactions. |
