川芎治疗偏头痛有效部位筛选研究

目的:研究川芎提取物对硝酸甘油所致偏头痛大鼠行为学症状及血浆、脑组织中的5-羟色胺(5-HT)、多巴胺(DA)、去甲肾上腺素(NE)含量的影响,初步筛选川芎治疗偏头痛的有效部位。方法:将川芎乙醇总提取物,采用系统溶剂法依次以氯仿、乙酸乙酯、正丁醇萃取,得到各提取部位后,分别灌胃给予实验大鼠。采用硝酸甘油致大鼠偏头痛模型,观察给药后大鼠耳红、挠头和爬笼次数等行为学指标;高效液相色谱-荧光检测法测定各组大鼠血浆及脑组织中的5-HT、DA、NE含量。结果:川芎乙醇提取物、乙酸乙酯提取物能显著抑制偏头痛大鼠耳红消失时间和挠头、爬笼次数;并能不同程度升高偏头痛大鼠血及脑内5-HT、DA、NE含量(P<0.05、0.01),且两者间无显著性差异。结论:乙酸乙酯提取部位是川芎治疗偏头痛的有效部位。

Preliminary Study on Effective Parts of Ligusticum Chuanxiong Against Migraine

Objective： To investigate the effect of Ligusticum Chuanxiong extract on behavioral indicators and 5-hydroxytryptamine（ 5- HT）,dopamine（ DA） and norepinephrine（ NE） contents in migraine rats,and to illustrate the effective parts. Methods： The experimental drugs were extracted by solutions method（ chloroform,ethyl acetate,N- butanol in turn） from ethanol extracts according to solutions（ polarity. SD rats were randomly divided into several groups and administrated with Ligusticum Chuanxiong extracts,and then the rat model of migraine was established by nitroglycerin,red ears and scratching frequency of all the rats were considered as behavioral indicators. Moreover,blood and brain were taken to detect 5- HT,DA and NE by HPLC- FLD method. Results： Ethanol extracts and ethyl acetate extracts of Ligusticum Chuanxiong had significantly reduced the disappearance time of model rats red ear,decreased the number of scratching their heads and climbed the cage of model rats,and significantly increased 5- HT,DA and NE levels of blood and brain tissue（ P〈0. 05,P〈0. 01） and there was no significant difference between the two extracts. Conclusion： Ethyl acetate extract was the effective parts of Ligusticum Chuanxiong in treatment of migraine.
