补阳还五汤含药血清对体外培养大鼠皮层神经元缺氧凋亡的影响

摘    要:[目的]探讨补阳还五汤对神经元缺氧凋亡的作用,以及对神经元缺氧过程氧自由基(oxygen free radicals,OFR)、一氧化氮(NO)生成和bcl-2基因表达的影响。[方法]采用Daniel方法建立神经元缺氧凋亡模型,加入补阳还五汤药物血清,应用磺化丙啶(propidium iodide,PI)染色法经流式细胞仪检测神经元凋亡率,分光光度法检测丙二醛(MDA)、NO浓度,免疫组化法检测bcl-2表达。[结果]补阳还五汤显著抑制缺氧导致的神经元凋亡,并减少神经元缺氧过程NO、OFR的生成,上调bcl-2基因的表达。[结论]补阳还五汤对神经元缺氧凋亡有抑制作用,其机制可能与其减少神经元缺氧过程NO、OFR生成,及上调bcl-2基因表达有关。

Abstract:To investigate the seropharmacological action of Buyang Huanwu Decoction (BHD) on hypoxia induced apoptosis of cultured cortical neurons. Models of hypoxia induced neuron apoptosis were established by Daniel method. Neuron apoptotic rate was examined by propidium iodide staining method and flow cytometry, nitric oxide (NO) and malonyl dialdehyde (MDA) contents by spectrophotometer and bcl 2 gene expression by immunohistochemical method. Effects of BHD on neuron apoptotic rate, NO and MDA contents and bcl 2 gene expression were observed in normal group (Group A), model group (Group B), normal saline group (Group C) and high dosage, moderate dosage and low dosage BHD groups (Group D, Group E and Group F). BHD could inhibit the apoptosis of cultured cortical neurons, reduce the production of NO and MDA and promote the expression of bcl 2 gene.[Conclusion]BHD has an inhibitory effect on hypoxia induced apoptosis of cultured cortical neurons and its mechanism may be concerned with the reduction of NO and MDA production and the increase of bcl 2 gene expression.
