不同剂量FK506对周围神经端侧吻合功能恢复的影响

摘    要:
目的探讨不同剂量FILS06对神经端侧吻合功能恢复的影响。方法30只Wistar大鼠取右侧腓总神经切断后,近端结扎并反转缝于邻近肌肉上,远端与胫神经外膜开窗处行端侧缝合。左侧为正常对照。术后分为5组,每组6只。于术后当天开始,每组于动物右小腿外侧肌肉内分别注射生理盐水(对照组)和生理盐水稀释的FKS06(实验组).剂量分别为lmg/kg-1·d~(-1)(实验组1)、2ms、kg~(-1)·d~(-1)(实验组2)、4mg/kg~(-1)·d~(-1)(实验组3)、8mg/kg~(-1)·d~(-1)(实验组4),连续2周。于术后3个月检测术肢双侧腓总神经、胫前肌电生理、组织学检查和胫前肌肌湿重的测定。结果实验各组全部电生理检测动作电位恢复率、肌肉单次收缩力恢复率、强直收缩力恢复率均显著高于对照组,再生神经纤维数目、肌湿重、肌纤维截面积均显著优于对照组。实验组2、3、4组之间差异无显著性意义.但均高于实验组1。结论FKS06对神经端侧吻合功能恢复具有促进作用。2mg/kg~(-1)·d~(-1)为比较合适的使用剂量。

Abstract:
Objective To study the effect of different dose of FK506 on peripheral nerve functional recovery after end-to-side coaptation.Methods 30 Wistar rats were used in this experiment.The peroneal nerve was transected on the right side of aLl the animals.The proximal end of peroneul nerve was turned back and ligated,while its distal end was sutured to the side of the tibial nerve with perineurial window.No operation was done on the left side. Then the animals were divided into 5 groups,with 6 each.Normal saline,FK506 2mg/kg/d,4mg/kg/d,8mg/kg/d was injected into the right tibial muscle in respectively according to group assignment once a day within two weeks.3 months after the operation,the peroneal nerve and the tibial muscle was harvested for electrophysiological and histologic examination,and the muscle wet weight was measured.Results (1)The FK506 groups had better results than the normal saline group.Significant difference was seen in the motor action potential,muscle contractility,number of regenerated nerve fibers,muscle wet weight and muscle fiber cross-sectional area.(2) No significant difference was found between the 2mg/kg/d,4mg/kg/d,8mg/kg/d FK506 groups.However the results were significantly better than those of the 1 mg/kg/d FK506 group.Conclusion FK506 can promote peripheral nerve functional recovery in end- to-side coaptstion.2mg/kg/d is a better dose to administer.
