膀胱上皮癌中细胞凋亡及相关基因C-myc的表达

摘    要:
[目的 ]探讨膀胱移行上皮癌的发病机制 .[方法 ]采用原位DNA片断末端标记及免疫组织化学技术 ,检测 42例膀胱移行上皮癌及 10例正常膀胱粘膜中的细胞凋亡及C myc蛋白的表达 .[结果 ]膀胱癌的细胞凋亡高于正常膀胱粘膜 ,但凋亡细胞计数随肿瘤的恶性转化而减少 .C myc阳性表达率为6 1 9% ,且低分化、浸润性膀胱癌C myc阳性表达率与高分化、浅表性膀胱癌比较有显著性差异 .C myc基因表达与病理分级及临床分期呈正相关 ,而C myc阳性组凋亡细胞计数与阴性组之间无显著性差异 .[结论 ]C myc蛋白参与了膀胱移行上皮癌组织中细胞凋亡的调节 ,并在膀胱移行上皮癌的发生和发展中发挥双向调节作用

Abstract:
OBJECTIVE*!To explore the mechanism of transitional cell carcinoma of bladder (TCCB).*!METHODS*!The apoptosis and the expression of apoptotic related gene C myc protein was detected by TDT mediated Dutp biotin nick end labeling(TUNEL) and immunohistochemistry methods in 42 case of TCCB and 10 case of normal mucosa of bladder.*!RESULTS*!The count of apoptosis of TCCB was higher than that of the normal control, but the count of apoptotic cells of TCCB decreased with increasing the malignant transformation. The positive rate of C myc expression was 61.9%, and there was a significant difference between the positive rate of C myc expression in poorly differentiated and infiltrating TCCB and well differentiated and superficial TCCB. The positive rate of C myc gene increased with the progression of the pathological grade and clinical stage, whereas there was no difference in the count of apoptosis between the positive group of C myc gene and the negative group of C myc gene.*!CONCLUSION*!The C myc gene takes part in the occurrence and progression of the TCCB, and the effect of C myc gene was on double aspects.
