小鼠单纯疱疹病毒I型潜伏感染模型的建立

摘    要:目的 建立简便易行的单纯疱疹病毒I型 (HSV_1)潜伏感染动物模型。方法 BALB c小鼠 6 0只随机分为 3组 ,分别为鼻黏膜病毒滴入组、尾静脉接种组和角膜划痕接种组 ,进行单纯疱疹病毒I型接种。 6周后行小鼠三叉神经节病毒抗原检测和病毒DNA片段检测 ,以确认潜伏感染的存在。结果 尾静脉接种组小鼠三叉神经节病毒DNA片段的检出率为 9 12 ,角膜划痕组小鼠三叉神经节病毒DNA片段的检出率为 13 16 ,均高于鼻黏膜接种 12 2 0 ;小鼠三叉神经节病毒抗原表达均为阴性 ;尾静脉接种组死亡率 4 15 ,高于其他两组。结论 鼻黏膜病毒滴入、尾静脉注射和角膜划痕接种均可建立小鼠HSV_1潜伏感染模型 ,以角膜划痕法引起疱疹病毒急性感染症状轻恢复快 ,潜伏感染发生率高 ,为最理想的建立HSV_1潜伏感染的方法。

Abstract:Objective To establish the convenient animal model of herpes simplex virus type-1(HSV-1) latent infection. Method BALB/c mice were divided into 3 groups: intravenous inoculation group,ocular abrasion group and intranasal inoculation group. Six weeks later the trigeminal ganglia(TG) were removed to detect the expression of HSV-1 antigen and the HSV DNA in TG was also detected by PCR to identify the existence of latent infection. Result The detected rate of HSV DNA in TG in intravenous inoculation group was 9/12 and was 13/16 in ocular abrasion group, which were higher than that in intranasal inoculation group(12/20). The expression of HSV antigen in TG in these 3 groups was all negative.The mortality of intravenous inoculation group was 4/15 which was much higher than those of two other groups. Conclusion The methods of intranasal virus dripping, cornea abrasion and intravenous injection can all establish the HSV-1 latent infection model in mice. Compared to two other groups cornea abrasion group shows less severe signs and a quicker recovery rate in acute infection and higher incidence rate of latent infection, so it is the ideal method to establish the HSV-1 latent infection model.
