狗牙花总生物碱的镇痛作用和精神依赖性的实验研究

目的 观察狗牙花总生物碱(Ervatamia yunnansis)的镇痛作用和精神依赖性.方法 选择昆明种雌性小鼠72只,其中小鼠热板法实验32只,条件性位置偏爱试验40只,按数字表法随机分为生理盐水对照组、吗啡组、狗牙花总生物碱低剂量组(5 mg/kg)及高剂量组(20 mg/kg)组.分别采用小鼠热板法和条件性位置偏爱试验观察狗牙花总生物碱镇痛作用和精神依赖性.结果 腹腔注射5、20 mg/kg狗牙花总生物碱后小鼠热板反应潜伏期显著延长(P<0.05或<0.01);狗牙花总生物碱5、20 mg/kg连续给药7 d后,小鼠在伴药侧逗留时间均无明显延长,没有形成条件性位置偏爱.结论 狗牙花总生物碱具有镇痛作用且无精神依赖性.

Experimental study on the effects of total Ervatamia yunnansis alkaloid on analgesia and psychological dependence

Objective To observe the effects of total Ervatamia yunnansis alkaloid on analgesia and psychological dependence in mice.Methods The hot plate test and conditional place preference test were used to valuate analgesic effect and the psychological dependence of total Ervatamia yunnansis alkaloid in mice.Results Following abdominal injection of total Ervatamia yunnansis alkaloid at dosages of 5 mg/kg and 20 mg/kg, latency of hot plate reaction of the mice was significantly prolonged(P<0.05 or <0.01).But, after medication of Ervatamia yunnansis at dosages of 5 and 20 mg/kg for 7 days, the time of staying in the drug-pair compartment wasn′t significantly extended and place preference wasn′t developed in mice.Conclusion Total Ervatamia yunnansis alkaloid seemed to have good analgesic effects and no psychological dependence was observed.
