摘    要:目的 建立液相色谱串联质谱(LC-MS)测定方法并检测犬以及人体埃博霉素B的血浆蛋白结合率.方法 建立LC-MS联合检测方法,以犬和人体血浆为对象,结合甲衡透析法检测埃博霉素B的血浆蛋白结合率.结果 3种浓度(100、50和10 ng/mL)埃博霉素B与犬血浆蛋白结合率分别为(85.4%±2.5%)、(81.3%±2.2%)和(83.7%±3 4%),与人体血浆蛋白结合率分别为(74.4%±3 8%)、(81.0%±2.9%)和(75.6%±2.2%),且灵敏度高,重现性好,操作简单.结论 埃博霉素B血浆蛋白结合率高,LC-MS可用于埃博霉素B血浆蛋白结合率的检测.

Abstract:Objective To establish the method with liquid chromatography-mass spectrophotometer (LC-MS) and determine the protein binding rates of epothilone B in plasma of dog and human. Methods The equilibrium dialysis combined with LC-MS was performed to determine the drug concentration of epothilone B, and its protein binding rates in dog and human plasma. Results With different concentration of epothilone B including 100, 50 and 10 ng/mL, the protein binding rates of epothilone B in dog were (85.4%±2.5%), (81.3%±2.2%) and (83.7%±3.4%), and in human were(74.4%± 3.8%), (81.0%± 2.9%) and (75.6%± 2.2%), respectively. Conclusion Epothilone B has higher level in binding to plasma protein of dog and human. The LC-MS method can be usd to determine the drug concentration of epothilone B properly.
