狭基线纹香茶菜水提部位对免疫肝损伤小鼠NF-kB蛋白表达水平及核转位的影响

摘    要:目的:研究狭基线纹香茶菜水提部位对免疫肝损伤小鼠核因子KB(NF—KB)蛋白表达水平及核转位的影响。方法:小鼠尾静脉注射刀豆蛋白A(ConA)25mg·kg。复制肝损伤模型,免疫组化法检测NF-kB在肝组织的表达,以及NF.KB从细胞浆向细胞核转位的情况。结果:复制模型4、8h后小鼠NF.KB蛋白表达显著增加(P〈0.01),吡咯烷二硫代氨基甲酸盐(PDTC)和狭基线纹香荼菜水提部位9.2g·kg。在4、8h均可显著降低模型小鼠NF.1(El的蛋白表达量(P〈O.01)。ConA能显著促进NF—KB自细胞浆向细胞核转位(P〈O.01)。PDTC和狭基线纹香茶菜提取物9.2g·kg-1。能显著减少NF-kB在细胞核表达,使其在细胞浆表达显著增多(P〈0.01)。结论:狭基线纹香茶菜可抑制免疫肝损伤小鼠NF-kB蛋白的表达,并抑制NF-kB的核转位。狭基线纹香茶菜对抗免疫肝损伤的作用机制可能与其抑制核NF-kB的激活有关。

Abstract:OBJECTIVE: To study the effect of water-soluble extract from Isodon lophanthoides on the expression of NF-kB protein and nuclear translocation in mice with immune hepatic injury. METHODS: Hepatic injury model was induced by tail intrave- nous injection of Con A 25 rag. kg-l. The expression of NF-~:B protein and nuclear translocation in liver tissue were detected by im- munohistochemistry examination. RESULTS: The expression of NF-kB protein increased significantly 4 h and 8 h after modeling (P〈0.01). PDTC and water-soluble extract (9.2 g·kg-1) can inhibit the expression of NF-kB protein(P〈0.01). ConA can improve that NF-KB translocated from cytoplasm into nucleus(P〈:0.01). PDTC and water-soluble extract (9.2 g. kg-1) reduced the expression of NF-kB protein in nucleus, and increased that in cytoplasm. CONCLUSION:1. lophanthoides can down-regulate the expres- sion of NF-kB protein and inhibit nuclear translocation. Inhibiting effect of 1. lophanthoides on immune hepatic injury is associated with the inhibition of the activation of NF-kB .
