摘    要:
目的大规模发酵、纯化幽门螺杆菌尿素酶B亚单位(ureB)基因疫苗,并对纯化后的疫苗进行鉴定。方法大规模发酵DH5α(pT ureB),收获的细菌经碱裂解后,用两步离子交换层析方法分离、纯化疫苗,通过紫外分光光度计、酶切、体外转染等对疫苗进行鉴定。结果发酵幽门螺杆菌ureB核酸疫苗,并经离子交换层析方法纯化获得了高质量的疫苗。结论纯化的ureBDNA疫苗为进一步的免疫实验奠定了基础。

Abstract:
Objective To ferment, purify and identify ureB gene vaccine of H. pylori. (Methods) Fermented DH5a (pT-ureB) were lysed using the alkaline method, and lysate was dealed with two-step anion-exchange chromatography. Then the purified gene vaccine was further analyzed by spectrophotometer, restriction enzyme digestion and transfection. Results The ureB gene vaccine were fermented successfully, and the purified gene vaccine was of high quality. Conclusions Purified ureB gene vaccine lays the foundations for further experiments in vivo.
