偏光显微镜在组织工程血管构建中的应用

摘    要:
目的利用偏光显微镜对组织工程构建血管壁中的胶原成分进行定性分析。方法体外培养、扩增的人脐带动脉平滑肌细胞,将其与可降解生物材料聚乙醇酸(PGA)制成的管状结构结合(实验组),体外培养1周后植入裸鼠背部皮下。于植入后2、4、5、11周时分别取材,普通光镜(HE和Masson染色)及偏光显微镜下观察组织学形态,并比较结果。以无细胞单纯管状PGA-DMEM植入裸鼠背部皮下作为对照。结果偏光显微镜下,随着植入时间的延长,实验组的PGA-平滑肌细胞复合结构从以PGA为主向以胶原和平滑肌细胞为主要成分过渡;对照组则随着PGA的降解无胶原成分形成。偏光显微镜检测结果与光镜下的组织形态学检查结果相同。结论偏光显微镜可用于组织工程血管壁中胶原成分的检测,方法简便且结果可靠。

Abstract:
Objective To qualitatively analyze the collagen in tissue engineered blood vessel under polarized microscope. MethodsTest group: smooth muscle cells were harvested from human umbilical artery,cultured and proliferated in vitro,and then were planted into a vessel made of degraded biomaterial PGA.One week later,the PGA-cells complex were implanted into nude mice under the back skin,and at week 2,4,5,6 and 11 after implantation,the complex were taken out for morphologic,historical(HE and Masson staining) and polarized microscope examination.Control group: only no-cell PGA vessel was implanted under the nude mice back skin,and the examination time was the same as test group. Results The polarized microscopy suggested that as time went on,the dominant component of the vessel changed progressly from PGA to collagen.The same finding was observed with the historical test.However,in the control group,as the PGA degraded,no collagen was formed.Conclusion It is a simple,convenient and reliable way to detect collagen in tissue engineered blood vessel with polarized microscope.
