摘 要: | 目的 :旨在探讨纹状体神经干细胞 (striatum neuralstemcells ,strNSCs)的培养及分化鉴定方法。方法 :选择性分离纹状体的神经干细胞 ,体外培养、扩增和诱导分化 ,并采用免疫荧光细胞化学检查鉴定。结果 :从胚鼠纹状体分离的细胞具有连续克隆能力 ,绝大多数细胞表达巢蛋白。诱导分化后的细胞表达成熟神经细胞和星形胶质细胞特异性的蛋白。结论 :用此方法分离的细胞具有自我更新、增殖和分化潜能 ,具备中枢神经系统干细胞的一般特征。

Cultivation and Differentiation of Embryonic Striatum-neural Stem Cells in Vitro |

Abstract: | Aim:To explore cultivation,differentiation and identification of embryonic striatum-neural stem cells ( str NSCs). Methods: str NSCs were isolated from the striatum of embryonic E14 rat and proliferated and differentiated. Immunoflurenscence cytochemistry were used for identifying them. Results:The str NSCs isolated from embryonic striatum had the potential to form clones, express neuroepithelial stem cell protein and differentiate into mature neurons and astrocytes. Conclusion: str NSCs isolated from embryonic striatum have NSCs's common features such as self-renewal, proliferation and pluripotentiality. |
