Study on the tumor-burdened SLID mice after umbilical cord blood transplantation immunity function changes

Objective To search for the ganulocyte-macrrophage colonyatimulating dry/progenitor cells graft reconstruction tumor-burdened SCID mice immunological mechanism for load whether people RenYuan sex tumour activated immune function.Methods firstly cultivate human liver cell HCCLM6 high transfer HCCLM6 lines,and then taking the cells in umbilical cord blood transplantation immunity function reconstruction group(group A) SCID mice subcutaneous establish tumor-burdened model,umbilical cord blood transplantation immunity function reconstruction group B not tumor-burdened as control.Experimental animals in five weeks after death,separately below detection.Compare A and B between groups immune function index variations,which flow cytometric analysis analysis mice with nuclei cells go-between peripheral blood lymphocyte proportion,ELISA determination people IgG content.The comparison group B 6 weeks and 11 weeks immune function indexes variation.Results 1 HCCLM6 lines inoculation in group A SCID mice subcutaneous tumor rate is 100 % into,into tumor incubation period for(9.83±1.47) d,After inoculation 5 weeks transplantation for(plus or minus 539.22 tumors secreting TSH 94.08) mm 3.5 weeks later SCID mice peripheral blood lymphocyte nuclear cells middleman CD3 CD4+ +,proportion/CD8+ respectively is: group A(1.50±0.40) %,Maurice;2.17±-Group B(2.17±0.50) %,1.19±0.66,two groups of comparisons,PCD3 = 0.04,PCD4 / CD8 = 0.08.People IgG content 48.57±respectively(4.51) muon g/ml,(61.68±its) muon g/ml,two groups of comparisons P< 0.01,there were significant differences.2.Group B lymphocytes proportion CD3+middleman,CD4±/ CD8±and person IgG content 6 weeks and 11 weeks,respectively(1.60±0.32) % and 2.36±0.74,(50.13±1.27) muon g/ml and(1.50±0.40) %,Maurice,(48.57 2.17± 4.51) muon g/ml,can see T lymphocytes in the first six weeks has reached a peak and can maintain to 11 weeks there is no obvious changes,and IgG content is have declined,but no statistically significant(P>0.05).Conclusion Tumor load conditions will influence on the immune function,namely,reconstruction of SCID mice immune activation RenYuan load people tumor thereby enhance its immure immune function,thus umbilical cord blood stem cell transplantation may be liver cancer treatment of a new method.
