燃煤型氟中毒对大鼠睾丸超微结构的影响

摘    要:目的研究燃煤型氟中毒对SD大鼠睾丸超微结构的影响。方法以SD大鼠为研究对象,按体质量均衡随机分为7组:对照组、高氟组、中氟组、低氟组、高氟加营养组、中氟加营养组、低氟加营养组。各染毒组喂饲含不同比例的燃煤型氟中毒病区煤烘玉米的饲料,复制燃煤型氟中毒动物模型。分3批(90 d,120 d,180 d)以股动脉放血法处死动物,查看氟斑牙,测尿氟。常规制作睾丸超薄切片,用透射电镜观察睾丸超微结构的变化。结果 (1)建成氟中毒动物模型,90 d各染毒组均出现氟中毒表现,对照组正常。中毒程度随氟剂量增加和染毒时间延长而加重;氟剂量相同时,营养好,中毒程度轻。(2)对照组结构正常。染毒组曲精小管内各级生精细胞生成减少,精子数量减少,各级生精细胞、睾丸间质细胞、肌样细胞、支持细胞的超微结构均有不同程度的病理变化。随氟剂量增加和染毒时间延长,病变加重。相同染氟剂量和加营养组比较,加营养组病变程度减轻。结论 (1)燃煤型氟中毒对SD大鼠睾丸组织超微结构有明显的损伤作用,随氟剂量和染毒时间延长,病变加重。(2)降低摄氟量及改善营养状况,可改善氟中毒对大鼠睾丸超微结构的损伤。

Abstract:ObjectiveTo study the effects of coal-burning fluorosis on testicular ultrastructure in SD rats.Methods SD rats taken as the experimental objects were randomly assorted into 7 groups which were control group,low-dose fluoride group,middle-dose fluoride group,high-dose fluoride group,low-dose fluoride supplemented with nutrition group,middle-dose fluoride supplemented with nutrition group and high-dose fluoride supplemented with nutrition group.The exposed groups were fed with the fodder with different proportions of corn dried by burning coal coming from endemic fluorosis areas,to establish the animal model of coal-burning fluorosis.All rats were killed by femoral arterial exsanguination at three times(90d,120d,180d),followed by examinations of dental fluorosis,fluorine in urine.Slides of testicular ultrathin sections were made by conventionally produced and observed ultrastructural changes of testis by transmission electron microscopy.Results Animal model of fluorosis was successfully established.There was fluorosis in every test group,but the rats in the control group kept norma1.The more serious fluorosis syndrome of the rats with higher the fluoride dose and change of time.When exposed to same fluoride dose,the milder their fluorosis syndrome with the better their nutrition.All the structure of the control groups kept normal.In exposed groups,the production of all levels of germ cells and spermatogenesis within the seminiferous tubules decreased.The ultrastructure of all levels of spermatogenic cells,Leydig cells,muscle cells and Sertoli cells had different degree of pathological changes.The lesions become more evident with the increase in fluoride dose and prolonged exposure.The less lesion,the better nutrition with the same fluoride dose.Conclusion Coal-burning fluorosis on testicular ultrastructure injury,with the fluoride dose and prolonged exposure,the lesions become more evident.Fluorosis can be resolved by decrease of fluoride intake and nutrition improvement.
