摘    要:
本文报道RU 486对离体人绒毛膜滋养层、蜕膜细胞的影响。并在小鼠胚胎与子宫内膜“共培养”模型上,观察不同浓度的RU 486对着床前、后胚胎及子宫内膜的作用。实验结果发现:当RU 486浓度为10~60μg/ml时,对绒毛滋养层细胞无直接影响;仅在浓度60μg/ml时,对蜕膜有轻度抑制作用。RU 486对小鼠胚胎及子宫内膜均有不同程度的影响,且随药物浓度的增加而作用增强。当浓度达10μg/ml时,几乎完全抑制胚胎的发育,并发生萎缩及退行性变化,同时子宫内膜也遭严重破坏。

Abstract:
This study presents the effects of RU486 on human chorio-trophoblast and decidual cells. We also have developed a tissue co-culture model of the embryo and endometrium for evaluating effects of RU486 on embryo development and implantation in vitro.The results showed that when 10~60 μg/ml of RU 486 were added to the treated culture media, they did not produce any significant change in trophoblast cell morphology as compared with the control group. However, when the concentration of RU 486 was 60 μg/ml, the growth of some decidual cells was slightly inhibited. In addition RU 486 also had the effects of inhibition on the endometrium and the embryos of mice in vitro. These effects became more prominent as the dose of the drug was increased. When the doses were increased to 10 or 20 μg/ml, the development of the embryes was inhibited and there was no embryo hatching from zona pellucida.. It was also noticed that atrophy and degeneration of embryos appeeared and the endometrium was injured at the same time.
