Abstract:
Objective To investigate whether abraxane regulates SLC25A25-AS1 to affect the proliferation,migration and invasion of lung cancer cell A549.Methods A549 cells were divided into control group,low-dose drug group,medium-dose drug group,high-dose drug group,high-dose drug+si-NC group,and high-dose drug+si-SLC25A25-AS1 group.Cell counting kit(CCK-8)method and colony formation experiment were performed to detect cell proliferation;Scratch assay and Transwell assays wad applied to detect cell migration and invasion;Western blot was used to analyze the expression levels of epithelial cell cadherin(E-cadherin)and neurocadherin(N-cadherin)protein.Results Compared with the control group,the cell viability,Clonal formation numbers,migration and invasion cell numbers,scratch healing rate and N-cadherin protein expression of A549 cells in the medium and high dose drug groups were significantly reduced(P<0.05),whereas the expression of E-cadherin protein and SLC25A25-AS1 were significantly increased(P<0.05).Compared with the high-dose drug+si-NC group,the cell viability,Clonal formation numbers,migration and invasion cell numbers,scratch healing rate and N-cadherin protein expression of A549 cells in high-dose drug+si-SLC25A25-AS1 group were significantly increased(P<0.05),whereas the expression of E-cadherin protein was significantly reduced(P<0.05).Conclusion Abraxane can inhibit the proliferation,migration and invasion of lung cancer cells A549,and the mechanism may be correlated to the up-regulation of SLC25A25-AS1 expression.
