| 摘 要： | 目的:探讨顺铂诱导的自噬在口腔鳞癌Tca83细胞中的作用。方法:利用顺铂作用于口腔鳞癌Tca83细胞,不同时间后MTT法检测细胞生存率的变化,激光共聚焦显微镜观察细胞胞浆中自噬体的形成情况,Annexin V-FITC流式细胞术检测细胞凋亡率的变化。结果:顺铂作用于Tca83细胞2h即可出现明显的自噬性变化。细胞凋亡率随顺铂作用时间的延长而逐渐增加。结论:顺铂可以诱导口腔鳞癌Tca83细胞发生自噬,过度激活的自噬作为细胞的一种死亡程序,可以与凋亡一起促进细胞死亡。

| Abstract: | Objective:To investigate the role of autophagy induced by cisplatin in oral squamous cell carcinoma Tca83 cell line.Methods:Being treated with cisplatin at different time,Tca83 cells survival rate was examined by tetrazolium bromide(MTT)colorimetry.Autophagosome formation was detected by Laser scanning confocal microscope.Apoptosis rate was checked by flow cytometry.Results:In the early(2h),autophagy was evident after the cells were treated with Cisplatin.Apoptosis rate increased with the extend time of cisplatin treatment.Conclusion: Cisplatin can induce autophagy in Tca83cell line,and overativation autophagy as a death procedure can promote cell death. |
