血管性痴呆大鼠海马神经元凋亡与乙酰胆碱含量变化

摘    要:目的 探讨血管性痴呆(VD)大鼠海马神经元凋亡和乙酰胆碱(ACh)含量变化在VD发病过程中的作用.方法 将90只大鼠随机分为模型组、假手术组和正常组,每组30只.再将各组的30只大鼠根据造模后断头取脑时间随机分为3小组,即7、14、21 d组,每组10只大鼠.采用反复大鼠双侧颈动脉缺血再灌注,加剪尾放血法制备VD大鼠模型;用TUNEL法检测海马组织的凋亡细胞,用生化法检测ACh含量.结果 模型组大鼠海马组织在第7、14、21天均有大量凋亡神经元,且ACh含量持续降低,与假手术组及正常组相比差异显著(P<0.01).结论 脑海马组织神经元的大量凋亡和ACh含量持续降低可能在VD发病过程中起重要作用.

Change of neuron apoptosis and acetylcholine in hippocampus of rat with vascular dementia
Abstract:Objective To investigate the change of neuron apoptosis and acetylcholine in hippocampus of rat with vascular dementia (VD). Methods 90 adult Wistar rats were randomly divided into model, sham-operation, control groups. Each group was randomly divided into three subgroups according to different time after being killed, including 7, 14 and 21 d after VD model. The imitative VD model was established by repeatedly clipping common carotid artery and bloodletting from rat′s tail. The cell apoptosis was tested with TUNEL technique, and the content of acetylcholine with immunohistochemistry in hippocampal. Results The apoptotic neuron of the hippocampal in the model group markedly increased and the acetylcholine decreased than those of the sham-operation and the control group at 7, 14, 21 days (P<0.01). Conclusions The increase of the neuron apoptosis and the decrease of the acetylcholine in hippocampal tissues may be the pathological reason of the VD.
