山茱萸多糖的提取及含量测定

摘    要:目的从山茱萸中提取多糖,并建立山茱萸多糖含量的测定方法。方法采用均匀设计优化山茱萸多糖的提取条件,用热水浸提、乙醇沉淀、DE-52柱层析的方法提取纯化山茱萸多糖,用苯酚-硫酸比色法测定多糖含量。结果山茱萸中多糖的含量为16.04%,平均回收率为99.01%,RSD=1.13%(n=3)。结论首次运用均匀设计优化山茱萸多糖的提取条件,提取得率高,苯酚-硫酸比色法测定山茱萸多糖含量稳定性、回收率较好。

Abstract:Objective To extract polysaccharide and establish the method for determination of the content of polysaccharides from Fructus Corni.Methods Uniform design test was used to extract polysaccharides from Fructus Corni.Polysaccharide was purified by water extraction,ethanol sedimentation,DE-52 chromatography.The content of polysaccharide was determined by phenol-vitriolic colorimetry.Results The content of polysaccharide in the Fructus Corni was 16.04%,the average recovery was 99.01%,and RSD was 1.13%(n=3).Conclusion It is the first time that uniform design is used to optimize extraction conditions of polysaccharide.The extraction rate of polysaccharide is high.Phenol-vitriolic colorimetry was a stable and reproduciable method to determine content of polysaccharide in Fructus Corni.
