高脂血症apoE基因敲除小鼠心脏中一氧化氮合酶含量及基因表达

摘    要:目的 探讨高脂饲料对apoE基因敲除(apoE-/-)小鼠心肌组织中内皮型一氧化氮合酶(endothelial nitric-oxide synthase,eNOS)的含量及基因表达的影响。方法 将13只apoE-/-小鼠随机分为两组,即高脂饲料组(n=7)和普通饲料组(n=6)。喂养12周后,测血清总胆固醇(total cholesterol,TC)、三酰甘油(triglyceride,TG)、高密度脂蛋白(high density lipoprotein,HDL)和低密度脂蛋白(low density lipoprotein,LDL)含量,采用Real-Time RT-PCR法检测心肌组织中eNOS的mRNA表达水平,以免疫组化方法检测eNOS蛋白的表达。结果 高脂饲料组TC、TG和LDL明显高于普通饲料组,HDL则低于普通饲料组(P<0.01);高脂饲料组心肌组织中eNOS的mRNA和蛋白水平均低于普通饲料组(P<0.01)。结论 高脂饲料导致apoE-/-小鼠发生高脂血症,并引起心肌组织中eNOS的mRNA表达的减少和相应蛋白水平的降低。

Abstract:Objective To explore the content and gene expression of endothelial nitric-oxide synthase (eNOS) in heart of hyperlipidemia apoE-/-mice.
