| 摘 要： | 目的 探讨GRIM-19 mRNA与蛋白的表达在结直肠癌发生、发展中的意义.方法 采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学方法检测40例结直肠癌患者癌组织,正常组织中的GRIM-19 mRNA及蛋白表达情况及测序分析.结果 结直肠癌组织中的GRIM-19 mRNA和蛋白水平明显低于正常组织,GRIM-19蛋白表达在癌组织中明显低于正常组织,且GRIM-19蛋白表达水平与结直肠癌分化程度和临床分期有关(P均＜0.05).结论 GRIM-19作为一种抑癌基因和分子标志物,其表达降低及失活可能与结直肠肿瘤的发生、发展有关.

| Abstract: | Objective To study the expression of grim-19 in colorectal carcinoma and its role in tumorigenesis.Methods The expression of grim-19 was detected by using RT-PCR and immunohistochemistry in the matched samples from 40 cases of malignant and normal tissues of colorectal tumors.Base sequences of the DNA product were determined.Resuits The expression level of grim-19 mRNA and protein in malignant colorectal tumors was obviously lower than in normal tissues.The expression of grim-19was correlated with clinical stage and cell difierentiation of eolorectal cancer ( P＜0.05 and P＜0.05 respectively).Condusion Grim-19 could be used as a novel tumor suppressor gene and molecular marker.Inactivation of grim-19 gene expression may be linked to oncogenesis. |
