软骨Ⅱ号方对兔骨关节炎软骨承受压缩载荷的影响

摘    要:目的:设计关节软骨的单向压缩实验,观察以中医学柔肝为治法的中药软骨Ⅱ号方对实验性家兔骨关节炎关节软骨生物力学性能的影响并探讨本方的作用机制。方法:按Hulth法建立家兔骨关节炎动物模型,并随机分为正常组、模型组、西药组(维骨力)、中药组(软骨Ⅱ号方)。用环氧树脂塑造与关节软骨表面紧密贴切的顶盖与底座,顶盖用以提供机器加载时的施力作用面并消除因关节软骨表面平坦度的差异所带来的随机误差,底座用以限制标本的滑动。测试前揩干软骨表面水份,用电子游标卡尺测算标本平均厚度为1.0mm,设定机器的恒定加载形变为0.5mm,恒定加载时间为2min,在AG-100KNA岛津万能试验机下进行单向压缩实验,同时自动描记形变-载荷曲线。结果:民空白组相比,模型组0.5mm形变对应的载荷值及硬度值明显降低(P<0.05),与模型组及西药组相比,中药组0.5mm形变对应的载荷值及硬度值明显升高(P<0.05)。结论:实验设计体现了关节软骨的生物力学性能,软骨Ⅱ号方明显提高了软骨抗压缩载荷的能力,改善了关节软骨的生物力学性能,其机制可能与改善胶原-蛋白多糖的结构有关。

Abstract:Objective: To observethe the effect of RGL on the biomechanical property of articular cartilage in osteoarthritic rabbits. Methods: The rabbits were divided into normal group, model group, Chinese herb group(RGL) and Viartril-s group, and osteoarthritic model were made according to Hulth's method .The cap and the bottom which were very suitable to the surface of the articular cartilage were made. The effect of the cap is to delete the random error because the surface of the articular cartilage is not smooth. The bottom is used to hold the moving of the specimen. Before the experiment, the water above the specimen's surface should be cleaned and the average thickness of cartilage was determined to be 0.5mm.The specimens were determined by the AG100KNA biomechal machine while design the same displacemant was 0.5mm and the same bear-time was 2 min. At the same time, the load-elongation curve was traced automatically. Result: The maximum load and stiffness is significantly different between the normal group and model group.(p<0.05) Comparing to the model group, the RGL group significantly improve the maximum load and stiffness.{p<0.05} Conclusion: The design of the experiment shows the biomechanical property of articular cartilage. RGL can improve the biomechanical property of articular cartilage and lt's mechanical property may relate to the construction of collagen and proteoglycans.
