化通脉注射液对人脐静脉血管内皮细胞ECV304增殖的影响

目的] 以血清药理学的方法观察血化瘀通脉注射液(HYTM)对于培养人脐静脉内皮细胞ECV304增殖的影响,为治疗性血管新生的靶基因选择提供线索。方法] 取对数生长期的内皮细胞,同步化于G0期,2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐(CCK-8)检测细胞增殖活性;酶联免疫吸附测定(ELISA)法检测5溴-2脱氧尿苷(BrdU)的掺入。结果] 与空白血清对照组相比,10%的HYTM含药血清体积添加分数可显著提高细胞增殖活性。结论] 10%的HYTM含药血清体积添加分数通过促进细胞DNA合成,可能具有保护血管内皮的作用。

Influence of Hua Yu Tong Mai injection on proliferation of ECV304 in human umbilical vein

Objective] To observe the influence of Hua Yu Tong Mai (Removing stasis and promoting circulation ,HYTM) Injection on the proliferation of ECV304 in human umbilical vein with the serum pharmacological method and provide a clue for choosing target gene for promoting vascular angiogenesis.Methods] The endothelial cell in logarithmic growth phase was obtained and synchronized in the G0 time.Their proliferation activity was examined by CCK-8; BrdU incorporate was examined with ELISA.Results] Compared with the blank blood serum control group,the volume increase score of medicine-containing serum with 10% HYTM.Conclusions] The volume increase score of serum containing medicine of 10% HYTM may promote endothelial cell proliferation through promoting DNA synthesis and mitosis and possibly has protective effect on vascular endothelium.
