| 摘 要： | 目的:比较3种兔骨髓间充质干细胞(BMSCs)分离方法的效率。方法:以12只3个月龄新西兰大白兔为实验对象,每只骨髓血平分3份,以全血细胞培养法、不连续Percoll分离液密度梯度离心法和Ficoll分离液密度梯度离心法进行分离,对克隆形成率、首次传代时间、16 d培养细胞数量等指标进行比较。结果:不连续Percoll分离液密度梯度离心法在各项指标中均优于Ficoll分离液密度梯度离心法及全血细胞培养法,且成功率最高。结论:不连续Percoll分离液密度梯度离心法是成年兔穿刺获取BMSCs最合适的分离方法。

| Abstract: | Objective:To compare the efficiency of three culture methods of mesenchymal stem cells derived from bone marrow(BMSCs)in rabbits.Methods:The bone marrow blood was taken from 12 Zelanian rabbits aged 3 months.BMSCs was cultured with the whole blood culture,the Percoll's discontinued density gradient solution and the Ficoll-Hypaque methods.The cloning formation rate,primary passage time and cell count after 16 days were observed.Results:For the three above-mentioned indexs,Percoll's discontinued density gradient solution was the best.Conclution:The Percoll's discontinued density gradient solution is the most preferable method for the isolation of BMSCs in adult rabbits. |
