摘    要:目的:探讨多巴胺受体(DRD4)基因第一外显子中NotI酶切位点甲基化与急性非洒巴细胞白血病(AML)的关系。方法:应用Southern印记杂交与DNA序列分析技术对AML患者骨髓标本进行分析。结果:在27例白血病患者骨髓标本中16例DRD4基因甲基化(59%),其中初治化疗敏感患者DRD4基因甲基化发生率为50%(9/18),复发及难治患者甲基化发生率为78%(7/9),而9例正常人骨髓标本DR

Abstract:Objective:To investigate aberrant DNA methylation of Not I site on exon I of dopamine D4 receptor (DRD4) gene in acute myeloid leukemia (AML).Methods:Southern Blot and DNA sequencing technique were performed on DNA from AML bone marrow samples. Results:Sixteen samples from 27 AML patients (59%) showed DNA methylation of DRD4 gene. Nine samples from 18 diagnostic chemosensitive patients (50%) and 7 samples from 9 refractory and relapse AML patients (78%) showed DNA methylation. But 9 normal bone marrow samples did not show any DNA methylation on DRD4 gene. Conclusion:The incidence of DRD4 gene methylation in AML was significantly higher than that in normal bone marrow ( P <0 05). There is potential importance of DRD4 gene methylation in initiation and development of AML.
