基于网络药理学探讨乌梅治疗哮喘的作用机制

摘    要:目的基于网络药理学方法,探讨乌梅治疗哮喘的作用机制。方法运用中药系统药理数据库和分析平台(TCMSP)检索乌梅的活性成分和靶标蛋白。运用治疗靶点数据库(TTD)检索哮喘的靶标蛋白。通过Cytoscape软件对二者的靶标蛋白进行交集,得到交集靶标蛋白并在Unitprot数据库查询交集靶标蛋白的基因名称。将交集靶标蛋白输入STRING数据库中构建蛋白相互作用网络。使用DAVID在线分析工具对交集靶标蛋白基因进行GO富集及KEGG通路富集分析。结果从乌梅中筛选出8种活性成分,涉及哮喘的靶标有14个。其治疗机制可能与神经活性配体-受体相互作用、钙信号通路、脂肪细胞脂解的调控、cAMP信号通路、利什曼病、胆碱能突触、TNF信号通路、弓形虫病有关。结论乌梅治疗哮喘涉及多种黄酮类、甾醇类化合物,可能与肾上腺素受体、肿瘤坏死因子、趋化因子、白三烯等有关,并涉及神经活性配体-受体相互作用、钙信号通路等通路。

Abstract:Objective To explore the mechanism of Wumei(Prunus mume)in the treatment of asthma based on network pharmacology.Methods The active ingredient and target protein of Wumei(Prunus mume)were searched using Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The target protein of asthma was searched using the Therapeutic Targets Database(TTD).Cytoscape software was used to intersect the target proteins of Wumei(Prunus mume)and asthma,and the gene names of the target proteins were queried in Unitprot database.The protein interaction network was constructed by inputting intersecting target proteins into STRING database.GO enrichment and KEGG pathway enrichment analysis of intersecting target protein genes were carried out by using DAVID online analysis tool.Results Eight active ingredients were screened from Wumei(Prunus mume),and 14 targets were involved in asthma.Its therapeutic mechanism may be related to neuroactive ligand-receptor interaction,calcium signaling pathway,regulation of lipolysis in adipocytes,c AMP signaling pathway,leishmaniasis,cholinergic synapse,TNF signaling pathway and toxoplasmosis.Conclusions The treatment of asthma by Wumei(Prunus mume)involves a variety of flavonoids and steroids,which may be related to adrenaline receptor,tumor necrosis factor,chemokine and leukotriene,and related to the pathways of neuroactive ligand-receptor interaction and calcium signaling pathway.
