柚皮素对脑缺血损伤小鼠神经细胞线粒体自噬的影响实验研究

摘    要:目的:探讨柚皮素对脑缺血小鼠神经细胞的保护作用,并从线粒体自噬途径探讨其保护作用的机制。方法:采用大脑中动脉栓塞造模(MCAO)的脑缺血再灌注损伤小鼠模型,原代皮层神经元的糖氧剥夺再灌注模型。动物实验分为空白组、假手术组、模型组、溶剂组、柚皮素组、抑制剂组、抑制剂+柚皮素组。细胞实验分为正常组、模型组、模型+柚皮素组、正常+柚皮素组、模型+抑制剂组、柚皮素+抑制剂组。检测神经功能缺损评分、脑梗死体积、细胞活力,观察小鼠脑组织神经细胞内粒体结构和线粒体自噬体分布情况,检测线粒体相关蛋白TOMM20、COX4I1、P62、LC3、Cyt C表达水平。结果:柚皮素可以降低MCAO小鼠的神经功能缺损评分,缩小脑梗死体积,增强神经细胞活力。电镜及Western blot结果发现柚皮素可以激活并促进MCAO小鼠神经细胞线粒体自噬。结论:柚皮素可以激活脑缺血后神经细胞线粒体自噬,减轻脑缺血后神经细胞损伤。

Abstract:Objective:Investigate the protective effect of naringenin on nerve cells in mice with cerebral ischemia injury,and to explore the mechanism of its protective effect from the mitophagy pathway.Methods:A mouse model of cerebral ischemia-reperfusion injury was established by middle cerebral artery embolization(MCAO),and a glucose-oxygen deprivation(OGD)-reperfusion model of primary cortical neurons.Animal experiments were divided into blank group,sham operation group,model group,solvent group,naringenin group,inhibitor group and inhibitor naringenin group.Cell experiments were divided into normal group,model group,model naringenin group,normal naringenin group,model inhibitor group and naringenin inhibitor group.The neurological deficit score,cerebral infarction volume and cell viability were measured,the structure of granulocytes and the distribution of mitochondrial autophagy in brain tissue of mice were observed,and the expression levels of mitochondrial related proteins TOMM20,COX4I1,P62,LC3 and Cyt C were detected.Results:Naringenin can reduce the neurological deficit score of MCAO mice,reduce the volume of cerebral infarction,and enhance the activity of nerve cells.Electron microscopy and Western blot showed that naringenin can activate and promote mitochondrial autophagy in MCAO mice and neurons.Conclusion:Naringenin can activate mitochondrial autophagy in nerve cells after cerebral ischemia and reduce nerve cell damage after cerebral ischemia.
