RP-HPLC法测定白头翁汤中白头翁皂苷B4的含量

目的 建立白头翁汤中白头翁皂苷B₄含量测定的RP-HPLC法。方法 采用Hypersil C₁₈色谱柱（250 mm×4.6 mm,5 μm）,以乙腈-水（体积比28∶72）为流动相,流速为1.0 mL·min^-1,检测波长205 nm,柱温30 ℃,进样量10 μL。结果 白头翁皂苷B4质量浓度在0.1～1.5 g·L^-1内线性关系良好（r=0.999 8）;平均加样回收率为103.1%,RSD为1.2%（n=9）;重复性RSD为1.6%（n=5）。结论 该方法简便、快速、准确、重现性好,为白头翁汤及其新剂型改进提供了一种质量控制方法。

Determination of pulchinenoside B4 in Pulsatillae decoction by RP-HPLC

Objective To establish an RP-HPLC method for the determination of pulchinenoside B₄ in Pulsatillae (traditional Chinese medicine) decoction. Method Chromatagraphy was performed on a Hypersil C18 column (250 mm× 4.6 mm, 5 µm) at 30 ℃ and 205 nm. The mobile phase was acetonitrile-water (V︰V= 28︰72,) at a flow rate of 1.0 mL·min^-1. Results The calibration curve was linear in the range of 0.1-1.5 g·L^-1（r=0.999 8）; The average recovery was 99.0% and RSD was 1.6% (n=9); The RSD of reproducibility was 1.6% (n=5). Conclusions The method is simple, accurate with good reproducibility. It can provide a method of quality assessment for Pulsatillae decoction.
