摘    要:目的 在大肠埃希菌中表达致密颗粒蛋白(GRA7)。方法 将弓形虫GRA7基因克隆入原核表达载体p GEX- 4 T- 1,构建重组质粒p GEX- 4 T- 1/GRA7并转化大肠埃希菌BL2 1。IPTG体外诱导重组质粒菌的表达,对表达的GRA7融合蛋白进行SDS- PAGE和Western- blotting分析。结果 成功构建了p GEX- 4 T- 1/GRA7重组质粒,该重组质粒在大肠埃希菌中表达了目的蛋白,该蛋白能与抗GST抗体结合。结论 GRA7基因在大肠埃希菌中以GST融合蛋白的形式得到表达。

Abstract:Objective To expressed GRA7 in Escherichia coli (E.coli).Methods GRA7 gene was cloned into the vector pGEX-4T-1.The recombinant plasmid pGEX-4T-1/ GRA7 was transformed into BL21 and induced by IPTG to express the fusion protein.This protein was identified by SDS-PAGE and Western-blotting.Results The recombinant plasmid pGEX-4T-1/GRA7 was constructed successfully and expressed the target protein in E.coli.This protein could react with anti-GST antibody.Conclusion The GRA7 gene may be expressed as a GST fusion protein in E.coli.
