白及多糖及其与5-Fu联用对人胃癌MKN45细胞株的影响

目的:探讨白及多糖及其与5-Fu联用对人胃癌MKN45细胞株的影响。方法:以人胃癌MKN45细胞株为对象,采用MTT法测定细胞增殖,HPLC测定细胞内药物浓度,对白及多糖及其与5-Fu联用对人胃癌MKN45细胞株增殖作用进行了观察研究。结果:白及多糖在体外无明显的直接杀伤人胃癌细胞作用,但低、中、高剂量的白及多糖与5-Fu联用均可增加5-Fu的抗肿瘤作用(P0.05);HPLC测定细胞内药物浓度结果显示,在5-Fu剂量相同的情况下,白及多糖与5-Fu联用可使细胞内5-Fu浓度增加(P0.05)。结论:白及多糖能增加5-Fu的抗肿瘤作用。

To investigate the effect of Rhizoma Bletillae polysaccharide and its combination with 5-Fu on human gastric cancer cell line MKN45

Objective： To investigate the effect of Rhizoma Bletillae polysaccharide and its combination with 5-Fu on human gastric cancer cell line MKN45. Methods ： Human gastric carcinoma cell line MKN45 as the object, cell proliferation was measured by MTT method, the intracellular drug concentration was determined by HPLC. Results： Rhizoma Bletillae polysaccharide in vitro without obvious direct killing effect on human gastric carcinoma cells, but low, high doses combined with 5-Fu can increase the antitumor effect of 5-Fu （P〈0.05）;HPLC mea- surement results showed that Rhizoma Bletillae polysaccharide can make the intracellular concentration of 5-Fu increased under the same conditions（P〈0.05 ）. Conclusion： Rhizoma Bletillae polysaccharide can enhance the antitumor effect of 5-Fu.
