大鼠脑缺血再灌注后NGF mRNA和NGFR的表达及其意义

摘    要:①目的 观察脑缺血再灌注对大鼠神经生长因子信使核糖核酸 (NGFmRNA)和神经生长因子受体(NGFR)表达的影响。②方法 线栓法制作大鼠脑缺血再灌注模型 ,采用原位杂交方法检测脑组织NGFmRNA的变化 ,免疫组织化学方法检测脑组织NGFR的变化。③结果 NGFmRNA在缺血周围区的表达 ,缺血再灌注 4h、1d、3d和 7d组较对照组明显增高 (t=9.3 2~ 13 9.5 0 ,P <0 .0 0 1) ;在缺血中心区 ,再灌注 4h组NGFmRNA表达也明显增高 (t=3 3 .0 9,P <0 .0 0 1)。NGFR在缺血周围区的表达 ,缺血再灌注 4h、1d、3d和 7d组较对照组明显增高 (t =17.2 8~ 40 .83 ,P <0 .0 0 1) ;在缺血中心区 ,再灌注 4h组较对照组明显增高 (t=13 .5 6,P <0 .0 1)。④结论 NGF与NGFR表达参与缺血再灌注后脑组织的保护作用

Abstract:Objective To study the expression of NGF mRNA and NGFR in neurons after reperfusion for focal cerebral ischemia in rats. Methods Zea Longa's model of reperfusion for ischemia was used for the study. The expressions of NGF mRNA were detected with in-situ hybrization and NGFR by immunohistochemical assay. Results In the peripheral area of the infarction, NGF mRNA positive neurons increased significantly at 4 hours, 1 day, 3 days and 7 days after reperfusion as compared with the controls (t=9.32-139.50, P<0.001); and in the central area, NGF mRNA positive neurons increased significantly only at 4 hours after reperfusion (t=33.09, P<0.001). NGFR positive neurons in the peripheral area of the infarction increased significantly at 4 hours, 1 day, 3 days and 7 days after reperfusion (t=17.28-40.83, P<0.001), while in the central area ,NGFR positive neurons increased significantly at 4 hours after reperfusion (t=13.56, P<0.01). Conclusion The expressions of NGF mRNA and NGFR are related to the protection of brain tissue of reperfusion for cerabral ischemia.
