摘    要: 目的研究微波辅助提取白芍中芍药苷的提取工艺和高效液相色谱测定芍药苷的含量的方法。方法采用均匀设计方法、微波提取和高效液相色谱测定,分别考察微波功率、提取时间、溶剂的浓度和料液比的选择对提取芍药苷的影响。结果测定的流动相为甲醇-0.05mol·L-1磷酸二氢钾-醋酸-异丙醇(67:173:4:4),流速为1.0mL·min-1

Abstract:OBJECTIVE To study the optimum condition for paeoniflorin in Paeonia lactiflora Pall extracted by microwave-assisted extraction(MAE) and to develop a HPLC method for determining the content of paeoniflorin.METHODS Using uniform design,MAE and HPLC,the effects of single parameter such as the microwave power,the radiation time,the solvent consumption and ethanol concentration in microwave-assisted extraction process on the content of paeoniflorin in Paeonia lactiflora Pall were investigated.RESULTS The mobile phase consisted of methol -0.05 mol·L-1 monopotassium phosphate-acetic acid-isopropyl alcohol(67∶173∶4∶4) at the flow rate of 1.0 mL·min-1,and the detection wavelentth was 230 nm.The optimum condition was as follows:the microwave power was 850 W,the solvent was 90% ethanol,the proportion of raw material to solvent was 1∶12,the radiation time was 25 min.CONCLUSION The HPLC method for paeoniflorin was accuracy.MAE methods was better than the conventional extraction methods.
